Abstract Intracellular nucleic acid testing process includes the following steps: providing labeled specific nucleic acid probe; fixing cell to be tested and exposing the nucleic acid to be tested; hybridizing specific nucleic acid probe and fixed suspending cell; washing out uncombined nucleic acid probe; fluorescent cascade amplifying label carried by the nucleic acid probe hybridizing combined with intracellular nucleic acid, and testing the fluorrescence with flow cytometer to obtain test result. The method may be used in qualitative, quantitative, timing test of intracellular nucleic acid level through combining with cell type, and is especially suitable for multiple-probe and multichannel analysis. The present invention also provides the device for the method.
