Abstract A three-phase extraction one-step method for extracting and purifying penicillin includes the following steps: adding macromolecular polymer/salt or two kinds of macromolecular polymers to pencillillin fermenting liquor or its filtrate according to a certain proportion to form double water phases; then adding organic phase to form three phases; regulating pH value, stirring and centrifugation to implement the processes of extraction, purification and removing impurity in one step; so extracting penicillin into the organic phase and distributing colour matter, impurity and heteroprotein into the double water phase, effectively purifying organic phase, reducing emulsion extent, simplifying flocculation and deemulsification of penicillin extraction, freeze-dewatering and decolouring. This invention can utilize density gradient and polar gradient of multi-phase system and can implement directional separation of target product and impurity simultaneously.
